HGF — SERPINE1
Text-mined interactions from Literome
Wojta et al., Lab Invest 1999
:
Therefore, we propose a role for HGF/SF in wound repair in the skin :
HGF/SF -- produced by activated fibroblasts --
increases in keratinocytes the expression of
PAI-1 , which leads to increased matrix stability during the repair process and which could also limit activation of HGF/SF by proteases such as urokinase-type PA ( u-PA ) or tissue-type PA ( t-PA )
Tacchini et al., Exp Cell Res 2003
(Neoplasms) :
The following findings support this conclusion : ( 1 ) HGF enhanced the activity of a luciferase reporter construct under the control of multiple HIF-1 responsive elements (HRE) in HepG2 cells, and the cotransfection of the dominant negative for the beta-subunit ( ARNT ) prevented this increase ; ( 2 )
HGF activated uPA and
PAI-1 promoters through HIF-1 activity regulated by PI3K/JNK1 transducers, as demonstrated by cotransfection with the reporter gene promoters and the dominant negative for ARNT, p85 subunit of PI3K or JNK1 ; ( 3 ) hypoxia was additive to HGF in increasing reporter vector activities, but probably through different transduction pathways ; ( 4 ) JNK1 wild-type expression vector increased HIF-1alpha protein expression probably in a phosphorylated state and, thus, functional for transactivating activity ; and ( 5 ) c-Jun did not seem to be involved in the activation of the luciferase construct containing multiple HREs because it was not prevented by expression of TAM-67, which is the dominant negative mutant form for c-Jun
Wang et al., Hepatology 2005
(Cholestasis...) :
Furthermore, PAI-1-/- mice had higher levels of tPA activation and mature hepatocyte growth factor (HGF) after BDL than WT mice, suggesting that
PAI-1 effects on
HGF activation critically influence cholestatic liver injury
Imagawa et al., Arterioscler Thromb Vasc Biol 2006
(MAP Kinase Signaling System) :
HGF increased PAI-1 mRNA expression and
PAI-1 protein accumulation in the conditioned media of human liver derived HepG2 cells, and increased hepatic PAI-1 mRNA expression in vivo in mice ... Because USFs are involved in the regulation of carbohydrates and lipid metabolism,
HGF mediated
PAI-1 production may provide a novel link between atherothrombosis and metabolic derangements
Wojta et al., Blood 1994
(Carcinoma, Hepatocellular...) :
Because u-PA has been shown to activate latent HGF to the active form, it could furthermore be speculated that by upregulating
PAI-1 , which in turn could inhibit u-PA,
HGF might
regulate its own activation
